摘要

【背景介绍】内质网 (ER) 膜蛋白复合物亚基 10 (EMC10) 与肥胖有关。作者研究了 EMC10 的两种同工型,包括分泌型同工型(scEMC10)和 ER 膜结合同工型(mEMC10),在代谢功能障碍相关的脂肪肝病 (MASLD) 中的作用。

【研究方法】采用多种脂肪变性小鼠模型和 HepG2 细胞来研究 EMC10 在肝脏 PERK-eIF2α-ATF4 信号传导和肝脂肪变性调节中的作用。探讨scEMC10中和抗体对小鼠肝脂肪变性的治疗作用。

【研究结果】scEMC10 促进肝脏 PERK-eIF2α-ATF4信号传导的激活,而mEMC10 抑制。基因敲除加剧了小鼠的肝脏 ER 应激和脂肪变性,而mEMC10的肝脏过度表达则受到缺乏蛋氨酸和胆碱的饮食或衣霉素的挑战,这突显了mEMC10通过调节肝脏 ER 应激在 MASLD 中的直接抑制作用。scEMC10 的过度表达促进了脂肪肝小鼠的肝脂肪变性,而循环 scEMC10 的中和则阻止了其发生,这表明scEMC10在MASLD发展中的作用。

这些发现证明了 EMC10 在 MASLD 发病机制中具有新的异构体特异性作用,并通过基于抗体的中和确定了分泌的异构体作为 MASLD 的易处理治疗靶点。

For transfection of lentivirus, NMCMs or HL-1 cells were seed in 6 cm dishes (for immunoprecipitation), 6-well plates (for immunoblots) or 96-well plates (for immunoflurorences) before transfecting with GST-Rilp or Rab7Y183A lentivirus following protocols from Genomeditech Co., Ltd. (Shanghai, China).
